A new thermophilic aldolase has been added to the biocatalytic tool-box for C-C bond formation

We have describe the heterologous expression of the rhamnulose-1-phosphate aldolase from Thermotoga maritima (Rha-1PA Tm) and its functional characterization. This enzyme presents an optimum temperature at 95 ºC and furthermore, in contrast with other hyperthermophilic enzymes, it maintains a significant percentage of activity at room temperature. The enzyme displayed a great stability to extreme reaction conditions such as high temperatures (half-life of more than 3 h at 115 ºC) and the presence of different organic solvents. Besides, is a useful biocatalyst that is compatible with multi-enzyme systems for C-C bond formation involving mesophilic enzymes.
